A pharmaceutical substance has an expiry period, which regulators need proof of. The proof is provided by conducting stability testing in controlled environments. A pharmaceutical stability chamber is the device used to provide this environment and maintain it over a period of months or even years. This article provides guidance on the requirements of ICH guidelines and selection of stability test chambers.
A stability chamber is an enclosed box. A stability chamber is used for controlling temperature and relative humidity (RH). A stability chamber can be employed for studying the effect of temperature, humidity, and light on pharmaceutical substances/products.
The data from these studies are used to:
A reliable stability test chamber is therefore a regulatory requirement as much as a piece of lab equipment.
The International Council for Harmonisation (ICH) publishes the stability guidelines that most regulators follow, including CDSCO (India), the US FDA, EMA, and WHO. The key ones are:
|
Guideline |
Scope |

ICH Q1A(R2) |
Stability testing of new drug substances and products (core guideline). |

ICH Q1B |
Photostability testing |

ICH Q1C |

ICH Q1E |

ICH Q5C |

Intermediate testing is generally triggered when a "significant change" occurs during accelerated testing for a product stored at 25°C/60% RH.
ICH divides the world into climatic zones, and your target markets decide which conditions apply.
If you export your products to Zone IV regions, then most likely, you would have to conduct long-term stability tests at either 30°C/65% RH or 30°C/75% RH. Most pharmaceutical organizations tend to opt for a unit that combines multiple stability conditions.
Light can affect many drug products. According to ICH Q1B, it is necessary to subject the sample to no less than 1.2 million lumens of visible light and 200 watt-hours of near-UV radiation per square meter. The procedure should be conducted in a special photostability chamber, which is a completely separate unit compared to the conventional temperature and humidity stability chamber.

The shelf-life of the product will be determined through stability testing, which will tell you how long the product will stay in the market and under what conditions it needs to be kept. The stability study will be done according to the ICH guideline Q1A(R2); however, it all depends upon the reliability of the equipment used.
Selecting a stability test chamber is not only about considering cost alone. It is important to consider uniformity, humidity, alarm system, and backup system, as well as what information the manufacturer has supplied. It is through the use of an appropriate chamber, proper SOPs, and calibration/mapping methods that accurate stability data will be achieved. It is extremely important to ensure that you are always updated on the current ICH guidelines.
What is a stability chamber used for in the pharma industry?
The stability chamber allows for controlled temperature and humidity levels to enable the manufacturer to investigate any changes in the drug substance or product during a certain period of time. This information is essential in order to establish shelf life and storage conditions.
What should be the duration of the stability study?
At the time of submission of the study, at least 12 months of long-term stability and 6 months of accelerated stability studies should be provided.
What should be the qualification requirement for a stability test chamber?
Most commonly, in addition to temperature and humidity mapping of the chamber under empty and loaded conditions, a qualification of IQ, OQ, and PQ should be performed by pharmaceutical companies.
How often do I calibrate the stability chamber?
Well, that is determined by your SOPs, your risk assessment, and any regulations that you think you will be required to comply with. Most laboratories perform annual sensor calibration, while some perform it even more often.
Is it possible that one stability chamber satisfies all the requirements set forth by the ICH?
Though it is generally possible to program stability chambers for different conditions, each time only one can be used. If several tests are to be conducted simultaneously, several chambers will be needed.
